Yep, that workedI think that is your problem.
SATA ports 5/6 are disabled when socket 2 is used for m.2 drives. I'm not sure if the top or bottom socket is labelled as no.2, but I would suggest putting the m.2 drive in the higher socket and see if you have the same issue. I would've suggested plugging the SSD in sata 1 through 4 if they are available as that would have been the easier way to test.
Edit: just found online manual for your board and even more convinced that is the issue. If you check page 10 it confirms SATA ports 5/6 are disabled when lower m.2 socket is used
