Which is the best/most reliable program for this? I want to make sure my Defiance II is running cool enough when it arrives.

Not sure the most reliable but I have used speccy and hwmoniter

All these programs to is to query the hardware sensors, so accuracy and reliability is down to the temperature sensors on the board and in the chips. I too use HWMonitor (http://www.cpuid.com/softwares/hwmonitor.html) and Speccy (http://www.piriform.com/speccy).
